A norepinephrine reuptake inhibitor (NRI, NERI) or Noradrenaline reuptake inhibitor or adrenergic reuptake inhibitor (ARI), is a type of drug that acts as a reuptake inhibitor for the neurotransmitters norepinephrine (noradrenaline) and epinephrine (adrenaline) by blocking the action of the norepinephrine transporter (NET). Venlafaxine is the first antidepressant in a new drug class referred to as the serotonin noradrenergic reuptake inhibitors (SNaRIs). Nefazodone is a weaker serotonin and norepinephrine reuptake inhibitor, but a potent serotonin 5-HTreceptor antagonist.
Reboxetine is the only NARI available. It is effective in treating depression, and also seems to improve motivation, energy level and social functioning.
It is also the only tetracyclic antidepressant. Inhibiting the reuptake of norepinephrine tends to elicit stimulating effects, thereby increasing both cortical arousal and energy levels. For the antidepressants , classical tricyclic antidepressants were the most potent drugs at five of the seven receptors (all but alpha 2-adrenergic and 5-HT1A receptors). The chlorophenylpiperazine derivative antidepressants (etoperidone, nefazodone, trazodone) were the most potent antidepressants at alpha 2-adrenergic. These may also be called selective serotonin-norepinephrine reuptake Inhibitors (SSNRIs).
SNRIs increase levels of serotonin and norepinephrine in the brain by blocking or delaying their reuptake by nerves. In our study, reboxetine, a novel selective norepinephrine reuptake inhibitor (NARI) was ranked first for efficacy followed by paroxetine (SSRI), doxepin (TCA), duloxetine (serotonin and norepinephrine reuptake inhibitors (SNRI)), trazodone (serotonin receptor antagonists and serotonin reuptake inhibitors), nortriptyline (TCA), citalopram (SSRI), sertraline (SSRI), placebo, nefiracetam (nicotinic acetylcholine receptors (nAChR)), fluoxetine (SSRI).
Selective Norepinephrine Reuptake Inhibitors (selective NRIs) are a group of drugs that exert action primarily by the inhibition of norepinephrine reuptake at the norepinephrine transporter protein. The affinity of a drug for such a receptor can be expressed mathematically as the dissociation constant (K i). NET) and the dopamine transporter (DAT), respectively.
